<title>Elektrochemiczne narzędzia diagnostyczne w pomiarach gazowego nadtlenku wodoru</title>

Hydrogen peroxide is one of the most important reactive oxygen species and plays a significant role in numerous biological and chemical processes. In the human body, it participates in biosynthesis, cellular signaling, and immune defense mechanisms. However, excessive accumulation of hydrogen peroxide and its elevated concentration in exhaled breath have been associated with impaired physiological function and may serve as potential biomarkers of diseases, including asthma and lung cancer. Therefore, accurate determination of gaseous hydrogen peroxide is important for modern medical diagnostics and environmental monitoring.&#13;
This Ph.D. dissertation focuses on the development of new electrochemical methods for the detection of gaseous hydrogen peroxide and the investigation of factors influencing the analytical performance of the sensing systems. The research involved the elucidation of electrode processes associated with the electrochemical reduction of hydrogen peroxide in the presence of copper(II) ions acting as an electrocatalyst, evaluation of screen-printed electrodes and surface modification strategies, and investigation of the influence of supporting electrolyte composition on the properties of the developed sensors. A key element of the work was the development of a measuring system based on the three-phase boundary concept and the implementation of advanced electrochemical techniques to improve sensitivity.&#13;
The conducted research led to new strategies for the determination of gaseous hydrogen peroxide, offering high sensitivity, good repeatability, and the capability for real-time measurements. The findings contributed to a better understanding of the mechanisms of electrochemical hydrogen peroxide reduction and provided new approaches that may be applied in the design of sensitive gas sensors.; Nadtlenek wodoru należy do grupy reaktywnych form tlenu i odgrywa istotną rolę w wielu procesach biologicznych oraz chemicznych. W organizmie człowieka uczestniczy między innymi w procesach biosyntezy, przekazywaniu i regulacji sygnałów komórkowych, a także w mechanizmach odpowiedzi immunologicznej. Jednocześnie jego nadmierna akumulacja i podwyższone stężenie w wydychanym powietrzu mogą świadczyć o zaburzeniu prawidłowego funkcjonowania organizmu i stanowić potencjalny wskaźnik wielu chorób, w tym astmy i raka płuc. Z tego też względu dokładna kontrola jego stężenia w fazie gazowej stanowi bardzo ważny element współczesnej diagnostyki oraz analizy środowiska. &#13;
	Niniejsza rozprawa doktorska poświęcona jest opracowaniu nowych elektrochemicznych metod detekcji gazowego nadtlenku wodoru oraz określeniu czynników wpływających na właściwości analityczne projektowanych układów pomiarowych. Badania obejmowały poznanie mechanizmów procesów elektrodowych zachodzących podczas elektrochemicznej redukcji nadtlenku wodoru w obecności elektrokatalizatora w postaci jonów miedzi(II), ocenę możliwości wykorzystania elektrod sitodrukowanych wraz z modyfikatorami ich powierzchni aktywnej oraz określenie wpływu składu elektrolitu podstawowego na właściwości analityczne opracowanych sensorów. Kluczowym elementem pracy było również opracowanie układu pomiarowego wykorzystującego granicę trzech faz oraz zastosowanie zaawansowanych technik elektrochemicznych w celu zwiększenia czułości i poprawy parametrów detekcyjnych. &#13;
	Przeprowadzone badania doprowadziły do opracowania nowych strategii oznaczania gazowego nadtlenku wodoru, charakteryzujących się wysoką czułością, dobrą powtarzalnością oraz możliwością prowadzenia pomiarów w czasie rzeczywistym. Uzyskane w niniejszej pracy wyniki pozwoliły na lepsze poznanie mechanizmów elektrochemicznej redukcji nadtlenku wodoru oraz wskazały nowe rozwiązania, które mogą znaleźć zastosowanie w konstrukcji wysoko czułych sensorów gazowych.

Na przestrzeni ostatnich dekad obserwuje się wzrost znaczenia tzw. chorób cywilizacyjnych, w tym chorób nowotworowych, układu krążenia oraz neurodegeneracyjnych. Ich rozwój wiązany jest m.in. z zaburzeniami metabolizmu małocząsteczkowych związków siarki, które mogą nasilać stres oksydacyjny i zaburzać równowagę redoks organizmu. Z tego względu oznaczanie tioli oraz ich pochodnych w matrycach biologicznych, takich jak osocze i mocz, ma istotne znaczenie w monitorowaniu stanu zdrowia. Coraz większą uwagę poświęca się również analizie żywności, w tym owoców będących źródłem substancji bioaktywnych o właściwościach antyoksydacyjnych. Prawidłowo zbilansowana dieta może bowiem wspierać profilaktykę chorób cywilizacyjnych oraz ograniczać tempo ich rozwoju.&#13;
Analiza związków tiolowych wymaga stosowania czułych i wiarygodnych narzędzi analitycznych ze względu na ich niskie stężenia, wysoką reaktywność oraz podatność na przemiany oksydacyjno-redukcyjne. Jedną z podstawowych technik stosowanych w tym celu jest wysokosprawna chromatografia cieczowa (HPLC), która w połączeniu z detekcją UV lub spektrometrią mas (HPLC-MS/MS) umożliwia selektywne i czułe oznaczanie analitów w złożonych matrycach. Odpowiedni dobór warunków przygotowania próbek oraz parametrów detekcji ma przy tym zasadnicze znaczenie dla jakości uzyskiwanych wyników.&#13;
Celem niniejszej pracy doktorskiej było opracowanie metod analitycznych opartych na HPLC-UV oraz HPLC-MS/MS, przeznaczonych do oznaczania wybranych biologicznie istotnych związków siarki w płynach ustrojowych oraz tzw. superowocach. Szczególną uwagę poświęcono optymalizacji przygotowania próbek, dążąc do uproszczenia procedur, poprawy precyzji i dokładności oznaczeń, skrócenia czasu analiz oraz ograniczenia ich uciążliwości środowiskowej. Uwzględniono także właściwości fizykochemiczne analitów oraz specyfikę badanych matryc i technik pomiarowych. W rezultacie opracowano cztery metody umożliwiające oznaczanie małocząsteczkowych tioli i ich pochodnych w moczu, osoczu oraz owocach jagodowych uprawianych w Polsce.; Over recent decades, attention has been paid to lifestyle diseases, including cancer, cardiovascular disorders, and neurodegenerative diseases. Their development has been associated with disturbances in the metabolism of low-molecular-weight sulfur compounds, which may intensify oxidative stress and disrupt redox homeostasis. Therefore, determination of thiols and their derivatives in biological matrices such as plasma and urine is important for health monitoring. Growing interest is focused on food analysis, especially fruits as sources of bioactive compounds with antioxidant properties. A balanced diet may support the prevention of lifestyle diseases and help slow their progression.&#13;
Analysis of thiol compounds requires sensitive and reliable analytical tools because of their low concentrations, high reactivity, and susceptibility to oxidation-reduction transformations. High-performance liquid chromatography (HPLC) is one of the principal techniques used for this purpose. Combined with UV detection or mass spectrometry (HPLC-MS/MS), it enables selective and sensitive determination of analytes in complex matrices. Appropriate selection of sample preparation conditions and detection parameters is essential for obtaining reliable results.&#13;
The aim of this doctoral dissertation was to develop analytical methods based on HPLC-UV and HPLC-MS/MS for determining selected biologically relevant sulfur compounds in body fluids and so-called superfruits. Particular attention was devoted to optimizing sample preparation to simplify procedures, improve precision and accuracy, reduce analysis time, and limit environmental impact. The physicochemical properties of analytes and the characteristics of matrices and measurement techniques were also considered. This study presents the effect of UV on the physicochemical changes of polystyrene nanoparticles (PS-NPs) with a size of 44 nm after 2, 4, and 8 h of illumination, along with its impact on white blood cells. It was found that UV illumination reduced the diameter &#13;
of PS-NPs to 10.2 ± 0.4% and increased the zeta potential to 36 ± 3% after 8 h of illumination. Microscopy (SEM and AFM) measurements showed considerable changes in the shape and behaviour of PS-NPs, indicating reduced size and a high tendency to agglomerate when deposited on the substrate surface. Moreover, UV-Vis spectroscopy showed an increase in the intensity of the n-π* band, signifying oxidation of the nanomaterial. The chemical modifications significantly influenced the biological properties of the nanoparticles, altering the morphology of peripheral blood mononuclear cells (PBMCs) as indicated by changes in forward scatter (FSC) and side scatter (SSC) parameters. Additionally, there were modifications in reactive oxygen species (ROS) generation, which, in turn, affected cell viability. Prolonged UV exposure of nanoparticles can also lead to increase in the levels of intracellular chelatable iron ions (Fe²⁺), thereby indicating the induction of cell death associated with ferroptosis. The most pronounced alterations in these parameters were observed after incubating PBMCs with nanoparticles exposed to UV for 8 hours.&#13;
Results from this study clearly demonstrate that the tested nanoplastic undergoes physicochemical changes upon exposure to UV, and that these induced changes strongly affect human peripheral blood mononuclear cells.
